It needs to be fuel resistant, and good to about 500 degrees, and doesn't set up. Gasket dressing, and or flange sealant.

The hylomar, and the permashield, will flow and fill

Well, totally sealed all the cracks (only tiny ones) and used sealant on the mating surface. Put it back together and test drove. Same results. But it does let me cruise, just no sudden touch of the throttle or it barks and coughs.

I may try to limp down to meet the group with it at Lake Geneva (43 miles). Then, limp to GB where others may help fix. If I can't limp, I'll stop at home and grab my truck.

You might switch the diaphragm from the bone stocker over to your pumpless carb, and take the washer on the bottom of the other stack, and put it on your stock needle, so you have the plastic spacer and 2 shims under the clip on the stock needle. That'll richen the mid with the stock needle, and rule out any tears or problems with the diaphragm in the carb you've been working on.
Dump about 4 ounces of Seafoam in your tank, when you leave out.

Here's a list of things to think about....

1. The pilot jetsrus sent, fits, but isn't the same as the one you pulled out. Easy enough to compare the two if you haven't already. The only difference, would be the size of the orifice, 35, versus 37.5 As long as all the outsides look the same, you'd be fine. They sell another pilot that is similar. Might have been a mix up.
2. While working on the carb, something got in and is restricting the pilot circuit. The bike can get the fuel to idle and start reving, but it doesn't get what it needs from the pilot, because of the clog, to make the leap into the mid...if this is the issue, when you get on the road, you should be able to cheat your way over this with some airflow, and a bit if extra throttle. One of the memebrs got a speck of something in his pilot jet, and could ride, but he had the one bad spot about where you are.
3. The o-ring around the PMS is gone, or is no longer sealing well. Unless you removed the PMS while you were working on the carb, and it fell out, this shouldn't be a problem.
4. The diaphragm got damaged when you where removing and replacing it. Possible. If it's not sealed tight, then the vacuum that is supposed to develop above it, won't happen, and the slide won't rise.
The manifold is still not getting a good seal. Some have had to do this twice to get it right(it's just a thought, not criticism).

When I say 4th groove from the top(blunt end of the needle, it ended up in the 4th groove from the bottom(pointy end) instead. Just tossing that out there The closer to the pointy end the clip is, the richer the midrange. If the clip is to close to the top(blunt end), you won't get the fuel you need for the transition.

The other carb may have not worked, because..
There was a manifold leak.
The pilot circuit was restricted. May be why the carb got sold(I don't know the history of it)
The stock jetting won't work with your setup, and was too lean.
